The caster's level comes into affect when a character has Spell Resistance. Caster's have to "beat" the spell resistance a character has by rolling a D20 and adding their level. SR = 12 Caster rolls a 10 and is fourth . . . .character is in trouble! Anyone feel free to correct me if I am wrong! But ...

Spell DC on pen and paper, is base 10 + spell level + attribute mod + shool of speciality if applicapable.

There are also feats which can increase the DC.

So, I cleric/druid with wis 18 (+4) and casting a third lvl. spell . . .
10 + 3 + 4 = 17
